Listeria monocytogenes by Real-Time PCR

Methodology: Real-Time PCR

Specimen Required: Collect: OneSwab® (loose stool) OR Yellow top tube (ACD solution A)
Specimen Preparation: For OneSwab:
Step 1. Utilize the swab provided to obtain a sample of loose stool and insert into the vial. 
Step 2. Remove the swab and place into the vial. Break the shaft at molded break point and insert into transport medium. 
Step 3. To prevent leakage, be sure the swab fits into the vial prior to capping. Tightly cap the vial and label with a minimum of two patient identifiers such as name and date of birth. 
For Blood:
Step 1. Completely fill the tube whenever possible to eliminate dilution from the anticoagulant and ensure the proper blood-to-anticoagulant ratio. 
Step 2. Immediately after blood collection, invert the tube gently 8-10 times to ensure mixing of anticoagulant with blood to prevent clotting. 
Step 3. Do not shake tube. 
Step 4. Do not centrifuge specimen.
Storage/Transport Temperature: Room Temperature 
Stability: One Swab: Room Temperature 5 days 
                 Blood: Room Temperature: 48 hrs.
